Carboxypeptidase B2 isoform A (CPB2), also known as Thrombin-Activatable Fibrinolysis Inhibitor (TAFI) or Carboxypeptidase U (CPU), is a plasma carboxypeptidase that regulates fibrinolysis. Synthesized primarily in the liver and expressed by platelets, CPB2 is activated by thrombin/thrombomodulin or plasmin, converting it into its active form (TAFIa). TAFIa removes C-terminal lysine residues from partially degraded fibrin, inhibiting plasminogen binding and activation, thus stabilizing clots. CPB2 also converts complement component C5a into its desArg form, modulating the immune response. Dysregulation of CPB2 may impact thrombosis risk or bleeding disorders, and genetic polymorphisms can influence its function.
Hydrolyzes C-terminal lysine or arginine residues from peptides/proteins, inhibiting fibrinolysis and modulating inflammatory mediators.
