Carboxypeptidase D (CPD) is a zinc-dependent metallocarboxypeptidase primarily involved in the processing of peptide hormones and neuropeptides within the trans-Golgi network. It preferentially removes C-terminal arginine or lysine residues from peptide substrates, functioning optimally at pH 5–7. It is a type I transmembrane protein with a large luminal domain containing three homologous domains (I, II, III), a single transmembrane region, and a short cytoplasmic tail. CPD plays a role in the biosynthesis of neuropeptides and peptide hormones and is associated with diseases like hepatitis B infection and lupus erythematosus.
