The Carcinoembryonic antigen peptide presented on HLA-A2 refers to a tumor-associated epitope formed when peptides derived from the carcinoembryonic antigen are processed and presented on the cell surface by the MHC class I molecule HLA-A*02:01. This peptide:MHC complex is recognized by cytotoxic T lymphocytes, making it a target for immunotherapies such as TCR-engineered T cells and cancer vaccines. However, the immunogenicity of native CEA peptides is often low, requiring the design of high-affinity peptide analogs to improve therapeutic outcomes. The major therapeutic challenge is balancing anti-tumor efficacy with off-tumor toxicity, as CEA is also expressed on certain normal tissues.
