Overview

Cardiac mesoderm enhancer-associated non-coding RNA (CARMN) is an evolutionarily conserved long non-coding RNA, originally characterized as a super enhancer-associated transcript critical for cardiac precursor cell fate and the maintenance of vascular smooth muscle cell homeostasis. CARMN serves as the host gene for the microRNAs miR-143 and miR-145, key regulators in cardiovascular development and disease as well as tumor suppression. CARMN is primarily nuclear and modulates gene expression through interactions with transcription factors (e.g., REST, myocardin, SRF), regulation of chromatin states, and involvement in RNA crosstalk (acting as a ceRNA). Its dysregulation is implicated in cardiovascular disease progression (e.g., atherosclerosis, aneurysm, heart failure) and several cancers, making it of growing interest for therapeutic targeting and biomarker discovery.
