Overview

Carica papaya leaf extract (CPLE) is a complex botanical mixture used primarily for the treatment of thrombocytopenia associated with Dengue fever. It does not have a single established molecular target; instead, its therapeutic effects are attributed to a synergy of phytochemicals including carpaine, papain, quercetin, and various phenolic compounds. Research suggests that CPLE enhances platelet production by increasing the expression of Arachidonate 12-lipoxygenase (ALOX12) and the Platelet-activating factor receptor (PTAFR), which are critical in megakaryocytopoiesis. Beyond its effect on platelets, CPLE exhibits anti-inflammatory and antioxidant properties that help maintain membrane integrity of blood cells during viral infections. Because it is a multi-component extract, it is classified as a therapeutic agent rather than a discrete molecular target like a receptor or enzyme. Mechanism of action

CPLE is thought to act through multiple pathways rather than a single receptor. Key putative mechanisms include the up-regulation of the ALOX12 (Arachidonate 12-lipoxygenase) gene, which increases megakaryocyte production and platelet count, and the modulation of the PTAFR (Platelet-activating factor receptor) gene. Additionally, its constituent flavonoids and alkaloids are believed to stabilize red blood cell membranes against hemolysis and provide antioxidant protection to hematopoietic cells.
