Overview

The carotid artery vessel wall is a tissue structure, not a discrete molecular target for drug development. It consists of three histological layers: the tunica intima (inner layer, endothelium supported by elastic/collagenous layers), tunica media (middle layer, smooth muscle regulating vessel diameter, blood flow, blood pressure), and tunica adventitia (outer layer, collagenous/elastic tissues anchoring the artery). The carotid bifurcation region also contains specialized sensory structures: the carotid body (chemoreceptor, regulates respiratory rate based on O2, CO2, pH) and the carotid sinus (baroreceptor, maintains blood pressure by detecting arterial stretch). Clinically, the carotid vessel wall is significant in atherosclerotic disease, where plaque accumulation can exhibit high-risk features like soft plaque, calcification, ulceration, neovascularization, and inflammation, correlating with increased risk of cerebrovascular ischemia. Due to its nature as a complex tissue structure rather than a single molecular entity, it is considered an incorrect and too anatomically general designation for a drug target.
