Overview

The carrier protein–derived peptide-MHC class II complex is a molecular assembly formed on the surface of antigen-presenting cells (APCs) such as dendritic cells and B cells. It consists of a peptide fragment, typically 13–25 amino acids long, derived from a carrier protein (e.g., CRM197, Tetanus Toxoid, or a viral vector) bound within the groove of a Major Histocompatibility Complex (MHC) class II molecule. The primary biological function of this complex is to be recognized by the T-cell receptor (TCR) of CD4+ T helper cells, which subsequently orchestrate immune responses, including B-cell activation and antibody production. In the context of modern biotherapeutics, this complex is a critical target for inducing immunological tolerance. By delivering the carrier protein alongside tolerogenic adjuvants like rapamycin (e.g., Selecta Biosciences' ImmTOR platform), researchers aim to prevent the formation of anti-drug antibodies (ADAs) that otherwise limit the efficacy and safety of enzyme replacement therapies and gene therapies. Consequently, the pMHCII complex serves as the focal point for strategies designed to reprogram the immune system from a reactive state to a tolerant state toward specific therapeutic antigens.

Mechanism of action
