Overview

Cartilage intermediate layer protein 2 (CILP2) is a secreted extracellular matrix glycoprotein specifically localized to the intermediate and deep zones of articular and meniscal cartilage. It is closely related in structure to CILP1 and shares the ability to be proteolytically processed and glycosylated. CILP2 interacts with collagen VI microfibrils and may mediate matrix component interactions. During cartilage degeneration, such as in osteoarthritis, CILP2 expression and protein levels in articular cartilage decrease, distinguishing its role from the upregulated CILP1 in similar contexts. CILP2 is not a receptor, enzyme, or transporter, but may be considered a specialized extracellular matrix structural protein, with potential utility as a biomarker for cartilage disease[1][2]. There are no known drugs that target CILP2 directly, and it is not currently a therapeutic target.
