Overview

CASC3 exon junction complex subunit (CASC3, also known as MLN51, Barentsz, and metastatic lymph node gene 51 protein) is a core component of the exon junction complex (EJC), a multiprotein assembly deposited on spliced mRNAs in the nucleus. CASC3 contributes to post-transcriptional regulation by marking mRNAs for subsequent control mechanisms, chiefly nonsense-mediated mRNA decay (NMD), which eliminates transcripts containing premature termination codons. This protein binds RNA and interacts with other EJC core components; though it shuttles between the nucleus and cytoplasm, its main function is in the cytoplasm, helping to regulate mRNA turnover and translation. CASC3 is not considered a classical drug target or biomarker and is only indirectly associated with disease through its regulatory role in mRNA processing and decay[1][2][4][6][7].
