Casein kinase 1 (CK1) is a family of serine/threonine-selective protein kinases found in all eukaryotes. CK1 enzymes are monomeric and function as key regulators in numerous cellular signaling pathways, impacting processes such as cell cycle progression, apoptosis, DNA repair, circadian rhythm regulation, and more. Mammals possess at least six or seven CK1 isoforms: alpha (α), beta (β), gamma 1–3 (γ1–3), delta (δ), and epsilon (ε). CK1 preferentially phosphorylates serine/threonine residues on substrates that are either preceded by acidic amino acids or primed by prior phosphorylation at nearby sites. Alterations in expression or activity levels of various CKI isoforms are linked to cancer development/progression as well as neurodegenerative diseases, making it a potential therapeutic target.
