Casein kinase 1 epsilon (CK1ε) is a serine/threonine protein kinase and a member of the casein kinase I family. It plays critical roles in regulating various cellular processes, particularly those related to circadian rhythms and signal transduction pathways. CK1ε is a central component of the mammalian circadian clock, participating in the transcription–translation feedback loop (TTFL) that generates ~24-hour cycles of gene expression and protein activity. Beyond its role in circadian regulation, CK1ε has been implicated in DNA replication and repair, Wnt/β-catenin signaling pathway activation, and regulation of synaptic transmission. Due to its pivotal role in controlling circadian rhythms, CK1ε is considered an attractive target for drugs aimed at treating sleep disorders, jet lag, mood disorders linked to rhythm disruptions, as well as certain cancers where cell cycle control is affected by aberrant clock function.
