Overview

Casein kinase 1 gamma-1 (CSNK1G1) is a serine/threonine protein kinase belonging to the casein kinase 1 (CK1) family, which includes several isoforms in humans. It functions as an enzyme that phosphorylates a wide variety of protein substrates, including acidic proteins like caseins. CSNK1G1 participates in key cellular processes, notably cell cycle checkpoint arrest in response to DNA replication stress, regulation of Wnt signaling pathways, and modulation of fast synaptic transmission via glutamate signaling. It is implicated in the phosphorylation of Claspin, a critical mediator for DNA replication stress responses. Mutations in CSNK1G1 have been associated with rare forms of epilepsy and possibly other developmental and cardiovascular disorders. While the broader CK1 family is significant in multiple signaling and regulatory pathways, no approved clinical drugs specifically target CSNK1G1, though research into kinase inhibitors continues due to the family’s roles in cancer biology, circadian rhythm, and neurodevelopmental diseases.
