Casein kinase II (CK2) is a highly conserved, constitutively active serine/threonine protein kinase with broad roles in cellular regulation and disease. It typically exists as a heterotetramer composed of two catalytic α subunits (either α or α′ isoforms) and two regulatory β subunits. CK2 regulates numerous cellular processes, including cell cycle progression, DNA repair, stress signaling, and gene expression. It is essential for cell viability and is overexpressed or hyperactive in many cancers, making it a target for anti-cancer drug development. It is also a drug target in fungal infections such as Cryptococcus neoformans.
