Casein kinase I isoform delta (CSNK1D)

Casein kinase I isoform delta (CSNK1D) is a member of the highly conserved casein kinase 1 family of serine/threonine-specific protein kinases that regulates a wide array of cellular processes [2, 11]. It is a central component of the eukaryotic circadian clock, where it phosphorylates Period proteins (PER1, PER2, and PER3) to regulate their stability and nuclear translocation, thereby determining the timing of the sleep-wake cycle [4, 14]. Beyond chronobiology, CSNK1D modulates key signaling pathways such as Wnt/beta-catenin and Hedgehog, and influences the activity of the tumor suppressor p53 and the DNA damage response [5, 10, 11]. In disease states, CSNK1D is often overexpressed in various cancers, including hepatocellular carcinoma and breast cancer, promoting tumor cell proliferation and metastasis [6, 8]. It is also implicated in neurodegenerative disorders like Alzheimer's disease through the hyperphosphorylation of tau protein and is linked to familial advanced sleep phase syndrome via specific genetic mutations [1, 12, 16]. Therapeutic development focuses on small-molecule ATP-competitive inhibitors to treat oncology, sleep disorders, and neurodegeneration [5, 9, 12].
