Casein kinase II subunit beta (CSNK2B) is the regulatory subunit of the tetrameric serine/threonine-protein kinase CK2, a highly conserved and constitutively active enzyme complex composed of two catalytic (alpha/alpha-prime) and two regulatory (beta) subunits[1][2][3]. The beta subunit encoded by CSNK2B is essential for enzyme stability, substrate selection, and regulation of CK2 activity, though the catalytic subunits retain activity in its absence[1][2][3]. CK2, including its beta subunit, is involved in broad cellular processes such as cell proliferation, apoptosis, DNA repair, stress responses, and signal transduction across pathways like PI3K/Akt, NFκB, Wnt/β-catenin, and JAK/STAT[1][2][3]. CSNK2B is frequently altered or overexpressed in cancer and is implicated in neurodevelopmental and neurodegenerative diseases, as well as resistance to chemotherapy[1][3]. Although most pharmacological efforts focus on the catalytic subunits, CK2 beta remains a critical determinant of enzyme function and cellular effects[3].
