Cbl-b is an E3 ubiquitin-protein ligase that acts as a negative regulator of immune cell activation, particularly in T cells and B cells. It controls ligand-induced down-modulation of the T cell receptor (TCR) and regulates signaling through the B cell receptor (BCR). As an E3 ubiquitin ligase, it targets key signaling molecules for ubiquitination-mediated degradation or signal attenuation. Targeting/inhibiting Cbl-b has emerged as a promising strategy for cancer immunotherapy.
