Overview

Caspase-4 is a human enzyme belonging to the caspase family of cysteine-aspartic acid proteases. It functions as an effector in the non-canonical inflammasome pathway, mediating LPS-induced pyroptosis. Upon activation by cytosolic LPS, it cleaves gasdermin-D and processes pro-inflammatory cytokines. It plays a critical role in innate immunity against Gram-negative bacterial infections, but dysregulation can contribute to pathological inflammation. It is primarily localized to the endoplasmic reticulum membrane. Inhibitors such as indoprofen are under investigation.
