Caspase-6 is a cysteine-aspartic acid protease (caspase) enzyme that plays a central role in programmed cell death (apoptosis), immune responses, and homeostasis. It functions primarily as an executioner caspase during apoptosis but also has unique roles in innate immunity and neurodegenerative diseases. Specifically, it mediates inflammasome activation and promotes multiple programmed cell death pathways. Caspase-6 cleaves substrates like poly(ADP-ribose) polymerase and nuclear lamins, and is implicated in diseases such as Huntington’s and Alzheimer’s by cleaving HTT and APP, respectively. Its activity is tightly regulated by proteolytic processing and interactions within the caspase cascade.
