Overview

Caspase 8-associated protein 2 (CASP8AP2; also known as FLASH) is a nuclear protein involved in the regulation of apoptosis and cell cycle progression[1][3][6]. It acts as a component of the death-inducing signaling complex (DISC), interacting with Fas receptor, FADD, and caspase 8 to regulate Fas-mediated apoptosis via the extrinsic pathway[1][3]. CASP8AP2 is essential for the activation of caspase 8 in response to death receptor signaling and is required for histone gene transcription and S phase progression in the cell cycle[6]. Additionally, the protein has transcription corepressor activity, participates in SUMO polymer binding, and mediates TNF-alpha-induced activation of NF-kappa-B via TRAF2 binding, acting independently of NF-kappa-B in some contexts[2][3]. Mutations or dysregulation of CASP8AP2 are implicated in hematological cancers and developmental disorders[3]. CASP8AP2 is commonly referred to by alternative names such as FLASH, FLICE-associated huge protein, and RIP25, reflecting its discovery in different signaling and apoptotic contexts[1][3][6].
