Overview

Caspase recruitment domain-containing protein 11 (CARD11) is a large, multi-domain scaffold protein that belongs to the membrane-associated guanylate kinase (MAGUK) family. CARD11 is primarily expressed in hematopoietic cells, especially B and T lymphocytes, where it orchestrates crucial signaling pathways following antigen receptor engagement. Upon activation by kinases such as PKC, CARD11 undergoes conformational changes allowing it to recruit signaling partners BCL10 and MALT1—forming the CBM complex, which is essential for the activation of effector pathways like NF-κB, JNK, and mTOR. These signals direct lymphocyte activation, proliferation, differentiation, and survival, underpinning both normal and pathological adaptive immune responses. Mutations in CARD11 can lead to immunodeficiency, autoimmunity, lymphoproliferative disorders, and cancer, signifying its importance as a master regulator and a therapeutic target in immune-mediated diseases[1][2][3][4][5][6][7].
