KatG is a bifunctional heme-dependent enzyme exhibiting both catalase and peroxidase activities. It is widely distributed among bacteria and lower eukaryotes, with particular significance in Mycobacterium tuberculosis, where it plays a crucial role in oxidative stress defense and antibiotic activation. It activates the antituberculosis prodrug isoniazid (INH) via its peroxidase activity. Mutations in the katG gene are a major mechanism of resistance to INH.
KatG activates isoniazid (INH) via its peroxidase activity. The activated drug then targets mycolic acid synthesis in the bacterial cell wall.
