Catalytic antibody 5C8 is a monoclonal antibody, also known as an abzyme, that functions as an artificial enzyme. It was specifically elicited against a transition-state analogue to catalyze the 6-endo-tet cyclization of trans-epoxy alcohols, a reaction that is stereoelectronically disfavored according to Baldwin's rules and lacks a known natural enzyme counterpart. The antibody's active site features a catalytic dyad of AspH95 and HisL89, which facilitates general acid-base catalysis and provides electrostatic stabilization for the reaction's transition state. The antibody was originally generated in Mus musculus and has been extensively studied through X-ray crystallography to elucidate its binding and catalytic properties. While 5C8 is a landmark molecule in the study of antibody catalysis and protein engineering, it is not a therapeutic target in the traditional sense. It is primarily used as a structural and mechanistic model to understand how the immune system can be programmed to create catalysts with high regio- and enantioselectivity. It should not be confused with the anti-CD40L antibody 5C8 (Ruplizumab), which is a therapeutic agent for autoimmune disorders. There are no known drugs that target this molecule, nor is it used as a drug in clinical practice.
