Overview

Catechol O-methyltransferase domain-containing protein 1 (COMTD1) is a human mitochondrial membrane-associated enzyme predicted to possess S-adenosylmethionine-dependent methyltransferase activity[1][2][3]. It shares sequence and structural similarity with the catechol-O-methyltransferase family. COMTD1 is ubiquitously expressed, with highest levels in tissues such as the gall bladder, small intestine, parathyroid gland, and kidney[2]. Its biological function is not fully established in humans, but in animal models, it is associated with regulation of mitochondrial metabolism, modulation of the oxidative stress response in pigment cells, and the synthesis of pheomelanin (a form of melanin pigment)[2]. Inactivation of COMTD1 in animal models alters various mitochondrial metabolic pathways and reduces pheomelanin synthesis but does not show evidence of involvement in well-characterized human diseases or therapeutic targeting to date[2].
