Catenin alpha-2 (CTNNA2), also known as alpha-N-catenin, is a cytoplasmic protein predominantly expressed in the nervous system. It functions as a mechanosensing adaptor, linking cadherin-based cell adhesion complexes to the actin cytoskeleton, thus regulating cell-cell adhesion, neuronal migration, and neurite outgrowth. CTNNA2 is crucial for neural development, cortical neuronal migration, and synaptic remodeling. Loss-of-function mutations are associated with neurodevelopmental abnormalities and cortical dysplasia. Unlike some other catenin family members, CTNNA2 is not primarily recognized as a direct therapeutic target but is important for understanding cell adhesion and cytoskeletal regulation in health and disease[1][2][3][5]. Note: No approved drugs directly target Catenin alpha-2, and it is not currently classified as a major receptor, enzyme, transporter, or therapeutic target in clinical pharmacology[1][3].
