Overview

Catenin delta-2 (CTNND2) is an adhesive junction-associated protein belonging to the armadillo/beta-catenin superfamily, predominantly expressed in the nervous system—particularly the brain. It plays a crucial role in cell adhesion, neuronal migration, dendritic spine and synapse development, and intracellular signaling. CTNND2's normal function is essential for proper neurodevelopment, and alterations or deletions are linked to conditions such as Cri-du-chat syndrome, autism spectrum disorders, intellectual disability, schizophrenia, and Alzheimer’s disease. It possesses oncogenic properties, influencing cell proliferation, invasion, and migration in tumors such as prostate adenocarcinoma, glioma, and melanoma. Although delta-catenin is a promising therapeutic and diagnostic target, no approved drugs directly interact with it as of now. However, its role in several cancers and neuropsychiatric disorders makes it an important disease-associated biomarker, and any therapeutic intervention involving CTNND2 must carefully consider neurological and oncogenic safety concerns
