Overview

Catsper channel auxiliary subunit gamma (CATSPERG) is a single-pass transmembrane protein that is an essential auxiliary component of the sperm-specific CatSper calcium channel complex. CATSPERG, together with other auxiliary subunits, helps assemble, stabilize, and regulate the CatSper complex, which is uniquely arranged in nanodomains along the sperm flagellum where it mediates calcium influx required for sperm hyperactivation and male fertility. Loss of CATSPERG disrupts CatSper assembly or trafficking, leads to male infertility in animal models, and identifies it as a critical checkpoint protein for proper function of the CatSper channel complex. The CatSper channel, including the gamma subunit, represents a promising target for the development of non-hormonal male contraceptives and for the understanding and treatment of certain forms of male infertility.

Mechanism of action

Not established for any existing drug. Hypothetically, agents inhibiting the CatSper complex (including the gamma subunit) would disrupt calcium influx into sperm, block motility/hyperactivation, and induce male infertility. Conversely, enhancing CatSper function could treat certain infertility phenotypes.
