Overview

Cation channel sperm-associated auxiliary subunit theta (CATSPERQ), encoded by the TMEM249 gene, is an essential auxiliary component of the CatSper calcium channel complex embedded in sperm flagella. It serves as a structural scaffold required for the proper assembly, dimerization, and higher-order organization of the CatSper channel by stabilizing the pore-forming subunit CATSPER4 and potentially facilitating CatSper dimer formation. CATSPERQ specifically localizes to sperm flagella and is testis-specific. Its absence causes a complete loss of the CatSper channel from sperm, leading to defective sperm hyperactivation and male infertility. CATSPERQ is not currently known as a direct therapeutic target or biomarker, but mutations or deletions cause impaired sperm function and infertility in males[1][2][3].
