Overview

CBY1-interacting BAR domain-containing protein 2 (CIBAR2), also known as FAM92B, is a membrane-associated protein containing a BAR (Bin/Amphiphysin/Rvs)-domain. It is implicated in the regulation of cilium assembly (ciliogenesis) and is predicted to localize to cellular structures such as the centriole, ciliary basal body, and ciliary transition zone[5][7][10]. CIBAR2 interacts with CBY1 and is thought to facilitate ciliogenesis, likely by aiding the recruitment and fusion of endosomal vesicles at distal appendages during the early stages of ciliary formation[5][6]. Although structurally closely related to FAM92A (CIBAR1) and capable of forming homodimers and potentially heterodimers, the exact molecular mechanisms and broader cellular roles of CIBAR2 remain poorly defined[2]. Current data do not support CIBAR2 as a recognized therapeutic target or as having known pharmacological modulators. Familial and sequence similarity as well as functional analogies suggest a role in diseases linked to ciliary dysfunction, including inflammatory bowel disease and nephronophthisis[5]. There are no known interacting drugs, biomarker roles, or specific safety concerns reported for this protein to date.
