C-C motif chemokine receptor 6 (CCR6)

C-C motif chemokine receptor 6 (CCR6) is a G protein-coupled receptor (GPCR) predominantly expressed on immune cells such as Th17 cells, B-cells, dendritic cells, and some innate immune cells[1][4][2]. It primarily binds the chemokine ligand CCL20 and plays a crucial role in directing leukocyte migration during inflammation and immune surveillance. By mediating chemotaxis of Th17 and regulatory T cells (Treg), CCR6 is a pivotal node in the regulation of immune homeostasis and pathogenic inflammation[2][4][5]. The CCR6/CCL20 axis has been implicated in multiple diseases, including chronic inflammation, autoimmune conditions, cancer, and infection. Although no therapies targeting CCR6 are yet approved for clinical use, several experimental therapies (antibodies, small molecules) aim to modulate this receptor due to its central role in immune cell trafficking and pathogenesis of inflammatory diseases[2][6][7]. CCR6 is also known by the alternative designation CD196 and is encoded by the *CCR6* gene situated at chromosome 6q27[4].
