Overview

CC chemokine receptor type 3 is a membrane-bound G protein-coupled receptor primarily expressed on eosinophils, basophils, mast cells, and Th2 lymphocytes. It binds several chemokines—most notably the eotaxin family (CCL11, CCL24, CCL26), MCP-3 (CCL7), MCP-4 (CCL13), and RANTES (CCL5)—to mediate chemotaxis, activation, and accumulation of these cells in sites of allergic inflammation and parasitic infection. CCR3 is implicated in the pathophysiology of asthma, allergic rhinitis, and other chronic inflammatory diseases, as well as serving as a co-receptor for HIV-1 entry into susceptible cells. Despite significant pharmaceutical interest and many small-molecule antagonists developed, no CCR3-based drug has yet achieved clinical success.

Antagonism of CCR3 blocks its interaction with chemokine ligands (e.g., eotaxins, MCP-3, RANTES), leading to reduced recruitment and activation of eosinophils and other immune cells, with therapeutic potential in allergy and inflammatory diseases.
