Overview

CCDC93 (CCC complex scaffolding subunit CCDC93) is a scaffolding protein that forms part of the **CCC complex** (COMMD/CCDC22/CCDC93 complex), which is crucial for the regulation of endosomal trafficking and recycling of various cell surface proteins, including integrins, signaling receptors, and channels[3][5]. CCDC93, together with CCDC22 and COMMD proteins (COMMD1-10), is essential for the function and stability of the CCC complex, facilitating its physical connection to the Retriever and WASH complexes that coordinate sorting and actin dynamics in endosomal compartments[1][2][5]. The CCC complex prevents lysosomal degradation of cargos and supports their return to the plasma membrane, thus playing pivotal roles in homeostasis, copper metabolism (through ATP7A trafficking), and cilium and centrosome assembly[1][2][3][5]. Mutations or deficiencies in CCDC93 or other CCC complex components can lead to disorders such as Ritscher-Schinzel syndrome and perturbations of copper homeostasis[3]. CCDC93 itself is not a classical therapeutic target (e.g., receptor, enzyme, transporter), and there are currently no drugs or biomarkers directly associated with it[3].
