CCR4-NOT transcription complex subunit 11 (CNOT11) is a conserved, non-catalytic protein component of the multi-protein CCR4-NOT complex, which is a central regulator of eukaryotic mRNA deadenylation and turnover[1][2][3]. CNOT11 forms a structural module with CNOT10, and both interact with the N-terminus of the large scaffold subunit CNOT1[2][3]. The primary role of the CNOT10/CNOT11 module is architectural, stabilizing the CCR4-NOT complex and promoting its assembly; CNOT11 is not known to possess enzymatic activity or act in isolation as a druggable target[1][2][3]. CNOT11 contains several structural domains, including a C-terminal MIF4G-like “antenna” domain, which serves as a platform for recruiting proteins such as GGNBP2 that control processes including spermatogenesis and possibly cell proliferation[3]. While the deadenylase activity of the CCR4-NOT complex is central to post-transcriptional gene regulation, CNOT11 is not catalytically active itself, and there are currently no drugs or direct disease links attributed to CNOT11 in isolation[1][3].
