Overview

CCR4-NOT transcription complex subunit 2 (CNOT2) is a core protein component of the highly conserved eukaryotic CCR4-NOT multiprotein complex, which regulates gene expression at both transcriptional and post-transcriptional levels. CNOT2 lacks catalytic activity but is essential for the structural integrity and deadenylase activity of the complex, governing mRNA degradation, stability, and processing, and mediating transcriptional repression through recruitment of repressor complexes like SMRT/NCoR-HDAC3. It influences cell cycle progression, apoptosis (especially when depleted), maintenance of embryonic stem cell identity, and has been implicated in tumor progression and metastatic susceptibility, particularly in cancer biology. CNOT2 is not currently recognized as a direct therapeutic drug target but is of interest in mechanistic and disease research due to its central regulatory functions.
