Overview

CCR4-NOT transcription complex subunit 6 (CNOT6) is the catalytic component of the CCR4-NOT complex, a highly conserved multi-protein complex involved in global regulation of mRNA stability and gene expression[1][3][5]. CNOT6 encodes a poly(A)-specific ribonuclease with 3′–5′ exonuclease activity, preferentially targeting polyadenylated RNA substrates and driving mRNA deadenylation—a key step in initiating mRNA decay, modulating miRNA-mediated repression, and controlling gene expression during key cellular processes such as proliferation and senescence[1][2][5]. Alongside its structural paralog CNOT6L, CNOT6 is essential for the deadenylase module of the complex but with distinct tissue expression and functional roles[5]. In summary, CNOT6 is an enzymatic factor, not a classical therapeutic target (such as a receptor or transporter), but plays a central regulatory role in post-transcriptional gene regulation[1][3][5].
