CD137 ligand (CD137L), also known as 4-1BB ligand or TNFSF9, is a type II transmembrane glycoprotein belonging to the TNF superfamily. It functions as a co-stimulatory molecule in the immune system, primarily expressed on antigen-presenting cells (APCs) like monocytes, macrophages, dendritic cells, and activated B cells. Upon binding to its receptor CD137 (4-1BB) on T cells, it delivers co-stimulatory signals that enhance T cell proliferation, survival, cytokine production, and cytotoxic activity. CD137L can also transmit reverse signals into APCs, leading to their activation and maturation. Due to its role in regulating immune responses, CD137/CD137L interaction is a target for immunotherapy, particularly in cancer, where agonists or antagonists are being developed to modulate anti-tumor immunity or control autoimmunity and allograft rejection.
