CD160 molecule is a glycosylphosphatidylinositol-anchored cell surface glycoprotein and member of the immunoglobulin superfamily, primarily expressed on natural killer (NK) cells and cytotoxic CD8 T cells[3][7]. It functions as a modulatory receptor, capable of both activating and inhibiting immune cell functions depending on the molecular and cellular context[1][2][7]. CD160 binds broadly to classical and nonclassical MHC class I molecules, as well as the herpesvirus entry mediator (HVEM), mediating cytotoxicity, cytokine secretion, and metabolic activation in NK and T cells[4][6][7]. Aberrant or altered expression of CD160 is associated with several diseases, including chronic lymphocytic leukemia (CLL), solid cancers, infection (notably HIV), and impaired immune tolerance. Its dual signaling nature—stimulating cytokine production and cell survival, or contributing to T cell exhaustion—makes it a promising target and biomarker in immunotherapy, cancer, infection, and immune regulation[1][3][7].
