CD164 molecule, sialomucin (CD164)

Target
CD164
Molecular classification
Cell adhesion molecule, Transmembrane sialomucin, CD molecule (Cluster of Differentiation)
01

Overview

CD164 (Sialomucin core protein 24, Endolyn) is a type I transmembrane sialomucin and cell adhesion molecule that regulates the proliferation, adhesion, and migration of hematopoietic progenitor cells, and modulates muscle development through interactions with CXCR4 and cell-cell adhesion mechanisms. It is implicated in stem cell biology, cancer metastasis, and is used as a diagnostic and therapeutic biomarker in multiple disease contexts. CD164 is highly glycosylated, displaying carbohydrate-dependent epitopes, and may mediate key cell contact and recognition events via its extracellular domains. Aberrant expression or mutation is associated with malignancy and sensorineural hearing loss.

02

Mechanism of action

Antibodies targeting carbohydrate-dependent epitopes modulate cell adhesion and proliferation; interference with CXCR4-mediated migration and homing (mainly experimental/biotherapeutic).

03

Biological functions

Regulation of cell adhesionRegulation of cell proliferationRegulation of cell migrationMyoblast differentiation and fusion (myogenesis)Regulation of hematopoiesis (blood cell development)Interaction with chemokine receptor CXCR4 (niche homing, migration)
04

Disease associations

Cancer (promotes tumorigenicity, prostate cancer metastasis, Sezary syndrome)Deafness/autosomal dominant sensorineural hearing lossOther roles: potential biomarker in hematologic malignancies, regulator in stem cell migration and homing
05

Safety considerations

Off-target effects due to broad expression in tissues and involvement in essential cell adhesion/migration processes.Specificity challenges when targeting CD164 therapeutically.Potential impairment of hematopoiesis or muscle regeneration.
06

Interacting drugs

Monoclonal antibodies (experimental)
07

Biomarkers

Marker for hematopoietic stem and progenitor cells (HSPC)Elevated expression as a marker in certain cancers (Sezary syndrome, prostate cancer metastasis)
