Overview

CD177 molecule is a GPI-anchored cell surface glycoprotein selectively expressed on a subset of human neutrophils, functioning primarily in immune regulation through adhesion, migration, and activation processes. It directly interacts with platelet endothelial cell adhesion molecule-1 (PECAM-1) to manage neutrophil transmigration across endothelial barriers and is necessary for cell-surface localization of proteinase 3 — a crucial antigen in antibody-mediated vasculitis. CD177 is involved in pathological processes such as myeloproliferative disease (polycythemia rubra vera), autoimmune vasculitis, and transfusion-related complications. A key feature is its role as the target for alloantibodies leading to neonatal neutropenia and transfusion reactions, as well as its modulation by therapeutic antibodies affecting integrin activation and cell migration
