Overview

CD1a is a member of the CD1 family of transmembrane glycoproteins, which are structurally related to MHC Class I molecules but specialized in presenting lipid and glycolipid antigens to T-cells. It is highly expressed on cortical thymocytes and professional antigen-presenting cells, particularly Langerhans cells in the skin and dendritic cells in the thymus. In the thymus, CD1a plays a critical role in the selection and maturation of T-cells by presenting endogenous or exogenous lipid ligands. Clinically, CD1a is a primary diagnostic marker for Langerhans cell histiocytosis (LCH) and is expressed in certain T-cell malignancies, such as T-cell acute lymphoblastic leukemia (T-ALL). Therapeutic strategies targeting CD1a involve monoclonal antibodies designed to deplete malignant cells or modulate lipid-mediated immune responses in inflammatory conditions.
