Overview

The CD1d–glycolipid complex is a molecular assembly where the CD1d protein, a non-polymorphic MHC class I-like molecule, presents lipid antigens to invariant Natural Killer T (iNKT) cells (PMID: 17637671). The complex is specifically recognized by the semi-invariant T cell receptor (TCR) of iNKT cells, which in humans typically utilizes the Vα24-Jα18 chain (UniProt: P15813). Upon binding, iNKT cells rapidly secrete a broad spectrum of cytokines, including interferon-gamma (IFN-γ) and interleukin-4 (IL-4), thereby modulating both innate and adaptive immune branches (PMID: 21217761). This interaction is a key target in immunotherapy, with synthetic glycolipids like alpha-galactosylceramide (α-GalCer) being used to stimulate anti-tumor responses or act as vaccine adjuvants (PMID: 23543120). In clinical settings, drugs like ABX196 are being evaluated for their ability to activate this pathway to treat solid tumors and enhance vaccine efficacy (ClinicalTrials.gov: NCT03897543). However, challenges include the potential for iNKT cell anergy and the risk of systemic cytokine release syndrome (PMID: 11861604).
