Overview

The CD1d-glycolipid complex consists of the CD1d antigen-presenting molecule bound to glycolipid antigens (such as α-galactosylceramide and its analogs). CD1d is a non-polymorphic, MHC class I-like molecule that presents lipid and glycolipid antigens to semi-invariant T cell receptors on iNKT cells; this interaction initiates potent immune responses. The structure of both the glycolipid and the CD1d binding groove determines the stability, specificity, and cytokine polarization of iNKT cells. The complex is a validated therapeutic target in multiple contexts, including immuno-oncology, autoimmunity, and infectious disease, and is a focus for drug development and immunomodulation. Safety and efficacy hinge on controlling the cytokine milieu generated by iNKT cell activation, as different glycolipid ligands bias toward distinct inflammatory or suppressive profiles.
