Overview

The CD1d-lipid antigen complex is a molecular assembly consisting of the CD1d protein—a non-polymorphic MHC class I-like molecule—and a lipid or glycolipid ligand. This complex is specifically recognized by the semi-invariant T-cell receptor (TCR) of invariant Natural Killer T (iNKT) cells, a unique subset of T cells that bridge innate and adaptive immunity (Nature Reviews Immunology, 2007). Unlike conventional T cells that recognize peptide antigens, iNKT cells respond to lipid-based antigens such as alpha-galactosylceramide (α-GalCer) (Frontiers in Immunology, 2018). Upon recognition of the complex, iNKT cells rapidly produce a diverse array of cytokines, including IFN-γ and IL-4, which can activate other immune cells like natural killer (NK) cells, dendritic cells, and B cells. This mechanism makes the CD1d-lipid complex a significant therapeutic target for cancer immunotherapy and vaccine adjuvants, where synthetic agonists like ABX196 are used to stimulate potent anti-tumor responses (Journal of Clinical Oncology, 2020). However, therapeutic application is complicated by the risk of iNKT cell anergy following overstimulation and the potential for systemic cytokine release syndrome (Cell Reports, 2019).
