Overview

CD1d-presented glycolipid antigens are a class of antigens consisting of glycolipids that are loaded and presented on the surface of antigen-presenting cells by the non-classical MHC class I-like molecule CD1d[2][5][6]. These antigens are recognized primarily by invariant natural killer T (iNKT) cells through their semi-invariant T cell receptor (TCR), leading to rapid cytokine secretion and modulation of immune responses[3][4][6]. The most widely studied glycolipid antigen is α-galactosylceramide (α-GalCer), originally discovered as a potent activator of iNKT cells. These antigens can be derived from self, microbial, or synthetic sources, and their structural features—especially the sugar head group and ceramide backbone—influence their affinity for CD1d and their capacity to activate iNKT cells[1][4]. Through these interactions, CD1d-presented glycolipid antigens play important roles in host defense against infection, immunoregulation, tumor surveillance, and are being explored as targets for immunotherapy and vaccine adjuvants[6]. The precise manipulation of such antigens enables the design of agents to steer immune responses for therapeutic benefit[4][6].
