CD2 antigen (CD2) is a transmembrane glycoprotein found predominantly on the surface of almost all peripheral T lymphocytes and thymocytes[1][2][5]. It belongs to the immunoglobulin superfamily and acts primarily as a cell adhesion molecule and signal transducer, facilitating adhesion between T cells and antigen-presenting cells (APCs) via interaction with its primary ligand CD58 (LFA-3 in humans)[1][3][6][7]. CD2 plays a significant role in the formation and organization of the immunological synapse, enhancing T cell receptor (TCR) signaling and T cell activation[2][3][6]. It is also involved in rearrangement of the actin cytoskeleton and participates in costimulatory signaling crucial for effective immune responses[2][5]. CD2-targeting therapies (such as alefacept, siplizumab) aim to modulate T cell responses for conditions like autoimmune disease and transplantation, but are associated with safety concerns including immunosuppression and increased infection risk[2].
