CD200 is a cell surface glycoprotein that plays a key immunoregulatory role through its interaction with the inhibitory receptor CD200R. This interaction delivers an inhibitory signal, attenuating immune responses. It is widely expressed on various cell types, including myeloid and lymphoid lineage cells, epithelial cells, endothelial cells, fibroblasts, and neurons, and is also present on many cancer cell types. CD200 is involved in maintaining self-tolerance, preventing excessive inflammation, and regulating tissue homeostasis. Dysregulation of the CD200/CD200R pathway is implicated in autoimmune diseases, chronic inflammation, tumor immunity, and bone homeostasis. Therapeutic strategies are being explored to target this axis for enhancing anti-tumor immunity or reducing pathological inflammation/autoimmunity.
