CD276 (B7-H3 protein) is a type I transmembrane glycoprotein and an immune checkpoint molecule belonging to the B7 family. Highly overexpressed in a broad range of human solid tumors, CD276 serves as a distinguishing marker for cancer stem-like cells, supporting tumor cell proliferation, migration, metabolic reprogramming, and immune evasion. Its expression correlates with tumor progression, metastatic capability, and poor prognosis. Therapeutic strategies targeting CD276 include monoclonal antibodies, bispecific antibodies, and CAR-T cells, which aim to block its immune-inhibitory effects and directly target cancer stem cells. The incomplete understanding of its specific receptors and occasional expression in normal tissue pose ongoing therapeutic challenges.
