CD33 is a transmembrane receptor belonging to the SIGLEC family, predominantly expressed on myeloid cells. It functions as an inhibitory receptor by binding to sialic acid-bearing glycans and recruiting phosphatases via ITIM motifs, thereby dampening immune cell activation. Its high expression in AML makes it a target for antibody-based therapies and CAR-T cell therapy.
Anti-CD33 antibodies bind to CD33, leading to cell death through antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), or direct apoptosis induction. Antibody-drug conjugates deliver cytotoxic payload upon internalization. CAR-T cells expressing anti-CD33 chimeric antigen receptors specifically target and kill CD33-expressing cells.
