CD4-positive T cell activation via Ag85B antigen presentation refers to the process by which CD4+ T helper cells are specifically activated in response to the presentation of the Mycobacterium tuberculosis antigen 85B (Ag85B) by professional antigen-presenting cells (APCs). This mechanism is central to adaptive immunity against tuberculosis. APCs process Ag85B into peptides, load them onto MHC-II molecules, and present them to CD4+ T cells in lymph nodes. TCR recognition of the peptide-MHC-II complex, along with CD4 co-receptor binding and co-stimulatory signals, leads to T cell activation, proliferation, and differentiation into effector Th1 cells, producing IFN-γ, IL-2, and TNF-α, which are critical for macrophage activation and granuloma formation. Memory T cell formation also occurs.
