CD4-positive T lymphocyte activation refers to the process by which CD4+ T cells (also known as helper T cells) are stimulated to respond to antigens. This activation is a central event in adaptive immunity, enabling these cells to orchestrate immune responses against pathogens and regulate other immune cell functions. Activation requires three signals: TCR engagement with peptide-MHC-II, co-stimulation via CD28-CD80/86 interaction, and cytokine-mediated differentiation cues. Outcomes include proliferation, differentiation into effector subsets, and execution of effector functions such as B cell help and macrophage activation. Dysregulation of CD4+ T cell activation can lead to various diseases.
