Overview

CD40 is a cell-surface receptor that belongs to the tumor necrosis factor receptor (TNFR) superfamily. It plays a central role in regulating immune activation and mediating both adaptive and innate immune responses. CD40 is primarily expressed on antigen-presenting cells (APCs) such as dendritic cells, B cells, and macrophages, but it can also be found on non-hematopoietic cells including endothelial cells, smooth muscle cells, fibroblasts, epithelial cells, and various tumor types. Upon engagement with its ligand or agonistic antibodies, CD40 triggers intracellular signaling cascades involving TNF receptor-associated factors (TRAFs), leading to the activation of transcription factors such as NFκB.
